The purpose of CsCl-ethidium bromide density gradient centrifugation is to separate supercoiled (covalently closed circular) plasmid DNA from other components present in lysates of bacterial cells: linear DNA (chromosomal fragments), nicked plasmid DNA (which lacks supercoils), RNA, proteins, and carbohydrates.
